Multifactorial Integrative Network Inference of NeurodeGeneration (MINING)

Neurodegeneration impacts brain function years before symptoms, presenting both a challenge and an opportunity to understand, treat and ultimately prevent dementia. The challenge lies in understanding why certain individuals can remain functionally resilient to advancing neuropathology and atrophy, and what determines their eventual conversion to a symptomatic state. Conversely, the opportunity lies in identifying the drivers of disease progression at the stage when intervention would be most desirable and effective.

This endeavour requires identifying the factors that moderate large-scale neural dynamics, while considering genetics, lifestyle, and overall health, and their consequences for cognitive function. The primary focus of the work was on providing an understanding of latent disease and conversion, through the integration of multimodal quantitative measures of brain structure, function and connectomics as determinants of cognition. By leveraging data from cohorts with known genetic risk and employing multivariate imaging analysis, I studied the critical role of neural dynamics in supporting cognitive ability in the presence of progressive yet presymptomatic pathology.

Follow up after fellowship:

The Fellowship allowed me to develop a novel framework and advance multivariate methodologies aimed at determining the distinct contributions of brain structure and function that are linked to differences in cognitive abilities. This work enabled me to obtain Alzheimer’s Society Dementia Research Leader Fellowship, where my research will focus on studying the brain changes underlying functional resilience in relation to genetic, vascular and comorbidity factors.
